Lower uterine segment involvement is associated with adverse outcome in patients with stage i endometroid endometrial cancer: results of a multicenter study.
CONCLUSIONS: In patients with apparent stage I endometroid endometrial cancer, the presence of LUSI is a poor prognostic factor, associated with a significantly higher risk of distal recurrence and death. The presence of LUSI warrants consideration when deciding upon surgical staging or postoperative management. Researchers find no link between ivf and breast cancer
Fertility treatment does not increase a woman's risk of developing breast cancer, according to a study of more than 25,000 women in the Netherlands. The large study will help to reassure patients concerned that the powerful hormone doses that are part of fertility treatment might put them at risk of developing the disease in the future. At the beginning of an IVF treatment cycle, women are given hormone drugs to stimulate their ovaries to produce more eggs so that clinicians can produce fertilised embryos in vitro. These lead to large spikes in oestrogen levels that could promote the development of breast cancer, which is sensitive to the hormone. The study, carried out by Dr Alexandra van den Belt-Dusebout at the Netherlands Cancer Institute in Amsterdam, compared 18,970 women who had had a...

The miseries of allergies just may help prevent some cancers
There may be a silver -- and healthy -- lining to the miserable cloud of allergy symptoms: Sneezing, coughing, tearing and itching just may help prevent cancer -- especially colon, skin, bladder, mouth, throat, uterus and cervix, lung and gastrointestinal tract cancer, as per a new Cornell study. These cancers, interestingly, involve organs that "interface directly with the external environment," said Paul Sherman, Cornell professor of neurobiology and behavior, who led the study. He and his colleagues analyzed 646 studies on allergies and cancers published over the past 50 years, putting together "the most comprehensive database yet available" on allergies and cancers........ Dkk3, downregulated in cervical cancer, functions as a negative regulator of [beta]-catenin
The Wnt/[beta]-catenin signaling pathway is activated during the malignant transformation of keratinocytes that originate from the human uterine cervix. Dkk1, 2 and 4 have been shown to modulate the Wnt-induced stabilization of the [beta]-catenin signaling pathway. However, the function of Dkk3 in this pathway is unknown. Comparison of the Dkk3 gene expression profiles in cervical cancer and normal cervical tissue by cDNA microarray and subsequent real-time PCR revealed that the Dkk3 gene is frequently downregulated in the cancer. Methylation studies showed that the promoter of Dkk3 was methylated in cervical cancer cell lines and 22 (31.4%) of 70 cervical cancer tissue specimens. This promoter methylation was associated with reduced expression of Dkk3 mRNA in the paired normal and tumor t...

Overexpression of hmga2 in uterine leiomyomas points to its general role for the pathogenesis of the disease
An overexpression of HMGA2 is supposed to be a key event in the genesis of leiomyoma with chromosomal rearrangements affecting the region 12q14-15 targeting the HMGA2 gene, but gene expression data regarding differences between uterine leiomyomas with and those without 12q14-15 aberrations are insufficient. To address the question whether HMGA2 is only upregulated in the 12q14-15 subgroup, the expression of HMGA2 was analyzed in a comprehensive set of leiomyomas (n = 180) including tumors with 12q14-15 chromosomal aberrations (n = 13) and matching myometrial tissues (n = 51) by quantitative RT-PCR. The highest expression levels for HMGA2 were observed in tumors with rearrangements affecting the region 12q14-15, but although HMGA2 is expressed at lower levels in leiomyomas without such aber...<p>&nbsp;</p><p><b><i>MedWorm Sponsored Message:</i></b> Find out how you can <a href="http://www.medworm.com/rss/medicalsponsorship.php" target="_self">get your message across here</a> by sponsoring this MedWorm news feed.</p>
The cytosol activity of thymidine phosphorylase in endometrial cancer
Conclusions: The cytosol TP activity in endometrial cancer is significantly higher than in normal endometrium, with no relation as to the stage and grade of tumors, but correlates with the PD-ECGF/TP protein expression and MD may therefore be associated with favorable prognosis in patients treated with chemo- or radiotherapy after surgery. In silico analysis of protein neoplastic biomarkers for cervix and uterine cancer
Abstract&nbsp;&nbsp;Worldwide, cervical and uterine cancers are the most deadly cancers in women, with high prevalences, especially in developing countries. The Human Protein Atlas (HPA) portal was explored for proteins expressed in a tissue-or cervix and uterine cancer-specific manner. The group of proteins differentially expressed and with enhanced expression in the glandular and surface epithelial (squamous) cells retrieved from HPA were further explored using the Protein Information and Knowledge Extractor (PIKE) portal to compile biological information that is found in different databases, and repositories on the Internet. Thus, the lists of candidate proteins found in HPA, and PIKE portals may be used as a starting point for the discovery and validation of biomarkers for cervix...

Does the fimbria have an embryologic origin distinct from that of the rest of the fallopian tube?
Conclusion(s): The two cases of pure congenital fallopian tube atresia, the presence of fimbriae in patients with müllerian (uterine, cervical, and vaginal) agenesis, and the role of the fimbria in ovarian-like and peritoneal cancers, support a novel hypothesis that the fimbria of the fallopian tube may arise separately from the rest of the tube. A microrna encoded in a highly conserved part of the mammalian hmga2 gene.
A microRNA encoded in a highly conserved part of the mammalian HMGA2 gene. Cancer Genet Cytogenet. 2008 Nov;187(1):43-4 Authors: von Ahsen I, Nimzyk R, Klemke M, Bullerdiek J The high mobility group protein HMGA2 plays an important role as a chromatin component of stem cells and as a protein causally related to the development of a variety of benign tumors (e.g., uterine leiomyomas, lipomas, and pleomorphic adenomas of the salivary glands). Herein, the existence of a highly conserved region within intron 3 of HMGA2 encoding a microRNA is described. The co-expression with HMGA2 suggests that as an intronic microRNA, this microRNA may cooperate with HMGA2 in its physiological and/or aberrant functions. Dietary genistein negates the inhibitory effect of letrozole on the growth of aromatase-expressing estrogen-dependent human breast cancer cells (mcf-7ca) in vivo
In conclusion, dietary GEN increased the growth of MCF-7Ca tumors implanted in ovariectomized mice and could also negate the inhibitory effect of LET on MCF-7Ca tumor growth. These findings are significant because tumors, which express aromatase and synthesize estrogen, are good candidates for aromatase therapy dietary and GEN can reverse the inhibitory effect of LET on tumor growth and adversely impact breast cancer therapy. Caution is warranted for consumption of dietary GEN by postmenopausal women with estrogen-dependent breast cancer taking LET treatment. (Source: Carcinogenesis)

Cyclooxygenase-2 expression in squamous cell carcinoma of the uterine cervix is associated with lymph node metastasis.
CONCLUSIONS: In this study, COX-2 expression was associated with lymph node metastasis in cervical squamous cell carcinoma, but this was linked to the presence of LVSI or parametrial involvement. This suggests that COX-2 expression may enhance lymph node metastasis after LVSI occurs. If so, immunohistochemical staining for COX-2 may provide additional prognostic information in LVSI-positive cases, in particular in patients who do not receive postoperative adjuvant treatment. Sex hormone binding globulin and aging
Horm Metab ResDOI: 10.1055/s-0028-1093351AbstractNew and more active concepts of steroid binding globulin action are emerging from recent research. As a result, examination of steroid levels in aging humans and the role of steroid binding globulins need to be re-visited. This review will discuss the possibility that sex hormone binding globulin (SHBG) plays an active role in the aging process. It will discuss the changes in blood levels of SHBG in aging humans in association with sexual activity, prostate hypertrophy and cancer, uterine leiomyoma, breast cancer, obesity and particularly the relationship between SHBG and HDL-cholesterol, Alzheimer's disease, osteoporosis, and cardiovascular disease. Starting with the idea that SHBG is an active participant in steroid action demands a re-eva...<p>&nbsp;</p><p><b><i>MedWorm Sponsored Message:</i></b> Find out how you can <a href="http://www.medworm.com/rss/medicalsponsorship.php" target="_self">get your message across here</a> by sponsoring this MedWorm news feed.</p>
Complex organisation and structure of the ghrelin antisense strand gene ghrlos, a candidate non-coding rna gene
Conclusion: GHRLOS RNA transcripts display several distinctive features of non-coding (ncRNA) genes, including 5' capping, polyadenylation, extensive splicing and short open reading frames. The gene is also non-conserved, with differential and tissue-restricted expression. The overlapping genomic arrangement of GHRLOS with the ghrelin gene indicates that it is likely to have interesting regulatory and functional roles in the ghrelin axis. (Source: BioMed Central)

Microrna signature and regulatory functions in the endometrium during normal and disease states
Semin Reprod Med 2008; 26: 479-493DOI: 10.1055/s-0028-1096128ABSTRACTDuring the menstrual cycle, human endometrium undergoes extensive cyclic morphologic and biochemical modifications in preparation for embryo implantation. These processes are highly regulated by ovarian steroids and various locally expressed gene products and involve inflammatory reaction, apoptosis, cell proliferation, angiogenesis, differentiation (tissue formation), and tissue remodeling. MicroRNAs (miRNAs) have emerged as key regulators of gene expression, and their altered and/or aberrant expression has been associated with establishment and progression of various disorders, including tumorigenesis. This review highlights the endometrial expression of miRNAs and their potential regulatory functions under normal and p...

Prevalence of high-risk human papillomavirus type 16/18 infection among women with normal cytology: risk factor analysis and implications for screening and prophylaxis
Conclusions: Although the sample size was small, based on the observation that HR-HPV 16 and 18 contributed significantly to the overall HPV prevalence in our setting, we speculate that testing/prophylaxis for these prevalent high-risk types could perhaps make cervical cancer screening and preventive programmes cost-effective. Larger community-based studies on HPV prevalence and persistence are required to validate these findings before definitive recommendations can be made to the policy makers. Novel therapeutic strategy for uterine endometrial cancers
Abstract&nbsp;&nbsp; In general, tumors induce angiogenic factors specific to them, which leads to angiogenesis with tumor progression. However, angiogenesis in uterine endometrial cancers is complicated because the hormone dependency in their growth also modifies the angiogenic potential. Therefore, angiogenic potential in uterine endometrial cancers must be thoroughly analyzed. The upstream of the vascular endothelial growth factor (VEGF) gene conserves estrogen-responsive elements. Progesterone primed with estrogen induces thymidine phosphorylase (TP) in the uterine endometrium. Sex steroid-dependent VEGF and TP are highly expressed in early-stage and well-differentiated uterine endometrial cancers, and basic fibroblast growth factor (bFGF) is highly expressed in advanced and poorly...
Is electron beam intraoperative radiotherapy (eliot) safe in pregnant women with early breast cancer? in vivo dosimetry to assess fetal dose
Abstract&nbsp;&nbsp;Electron beam intraoperative radiotherapy (ELIOT) is a new technique permitting breast radiotherapy to be completed in a single session. Since ELIOT is associated with much reduced irradiation to nontarget tissues, we carried out a study on nonpregnant breast cancer patients to estimate doses to the uterus during ELIOT to better evaluate the possible use of ELIOT in pregnant breast cancer patients. We performed in vivo dosimetry with thermoluminescence radiation detectors (TLDs) in 15&nbsp;premenopausal patients receiving ELIOT to the breast (prescribed dose 21&nbsp;Gy) using two mobile linear accelerators. The TLDs were positioned subdiaphragmatically on the irradiated side, at the medial pubic position, and within the uterus. A shielding apron (2&nbsp;mm lead eq...

Birth control has long term effect on hormone exposure
Researchers at Wake Forest University School of Medicine may be one step closer to understanding why past oral contraceptive use dramatically lowers the risk of ovarian and uterine cancers later in life. While studying the effect of post-menopausal dietary soy consumption on estrogen metabolism in cynomolgus monkeys, Latanya M. Scott, Ph.D., discovered that monkeys who had been given birth control earlier in life had a reduced amount of estrogen excreted in their urine. The rate of port-site metastases after 2251 laparoscopic procedures in women with underlying malignant disease.
CONCLUSION: The rate of port-site tumor implantation after laparoscopic procedures in women with malignant disease is low and almost always occurs in the setting of synchronous, advanced intraabdominal or distant metastatic disease. The presence of port-site implantation is a surrogate for advanced disease and should not be used as an argument against laparoscopic surgery in gynecologic malignancies. Mutagenicity of tamoxifen dna adducts in human endometrial cells and in silico prediction of p53 mutation hotspots
Tamoxifen elevates the risk of endometrial tumours in women and -(N2-deoxyguanosinyl)-tamoxifen adducts are reportedly present in endometrial tissue of patients undergoing therapy. Given the widespread use of tamoxifen there is considerable interest in elucidating the mechanisms underlying treatment-associated cancer. Using a combined experimental and multivariate statistical approach we have examined the mutagenicity and potential consequences of adduct formation by reactive intermediates in target uterine cells. pSP189 plasmid containing the supF gene was incubated with -acetoxytamoxifen or 4-hydroxytamoxifen quinone methide (4-OHtamQM) to generate dG-N2-tamoxifen and dG-N2-4-hydroxytamoxifen, respectively. Vitamin d a key player in overall health of several body organs, says uc riverside biochemist
(University of California - Riverside) Vitamin D, once linked to only bone diseases, is now recognized as a major player in contributing to overall human health, emphasizes UC Riverside's Anthony Norman, an international expert on vitamin D. He lists 36 organ tissues in the body whose cells respond biologically to vitamin D. The list includes bone marrow, breast, colon, intestine, kidney, lung, prostate, retina, skin, stomach and the uterus. According to Norman, vitamin D deficiency can impact all 36 organs. A joinpoint regression analysis of long-term trends in cancer mortality in japan (1958-2004)
Cancer is one of the major targets of disease control programs in Japan. A Joinpoint regression model was used to analyze the long-term trends of mortality related to overall cancer and the 15 most common cancers based on published data from the National Vital Statistics of Japan between 1958 and 2004. Since 1996, a decline has been seen in overall cancer for both sexes in Japan. Most of the common sites, including cancers of the stomach, colon, liver, gallbladder and lung and leukemia in both sexes, cancer of esophagus in men and rectal and ovarian cancers in women showed a decreasing trend, and cancers of the rectum, pancreas, prostate and urinary bladder and malignant lymphoma in men and cancers of the esophagus and uterus in women leveled off during the most recent period. However, an ...

Role of diffusion-weighted imaging in the diagnosis of gynecological diseases
Abstract&nbsp;&nbsp;Recent technical advances in diffusion-weighted imaging (DWI) greatly enhanced the clinical value of magnetic resonance imaging (MRI) of the body. DWI can provide excellent tissue contrast based on molecular diffusion and may be able to demonstrate malignant tumors. Quantitative measurement of the apparent diffusion coefficient (ADC) may be valuable in distinguishing between malignant and benign lesions. We reviewed DWI and conventional MRI of the female pelvis to study the utility of DWI in patients with gynecological diseases. Although the ADC can help to differentiate between normal and cancerous tissue in the uterine cervix and endometrium, its utility may be limited by the large overlap of the uterine myometrium and ovaries. On the other hand, the ADC may be ...

We hypothesize that regulatory and cell cycle protein expression might be additionally modified in the cervix of HIV/HPV co-infected women. Methods: We analyzed the expression of Rb, p27, VEGF and Elf-1 transcriptor factor by immunohistochemistry in 163 paraffin-embeded cervical samples using Tissue Micro-Array (TMA) and correlated this to HIV-1 and HPV infection. Results: HIV/HPV co-infection was associated with a significant increase in expression (p (Source: BMC Cancer)
